OP-023 Oxidative Stress in Chronic Heart Failure and Elevated Programmed Cell Death




Objective


Heart Failure comprises huge medical attention causing distinctive mortality and morbidity especially in westernized societies with a great impact on health care envisioning. The present study aimed to evaluate total antioxidant and oxidant capacities and advanced oxidative protein product levels via well-accepted methods in serum samples of patients with chronic heart failure (CHF). We also aimed to determine apoptosis levels in peripheral blood lymphocytes acquired from adult CHF patients and their the relationships with oxidative stress.




Methods


To the study, 32 adult patients with CHF and 32 sex-matched, healthy control groups were included. Blood samples were obtained following an overnight fasting state, serum samples were separated by centrifugation, then stored at -80°C, and used for the analysis of the levels of total antioxidant status (TAS; in μmol Trolox Eq/L), total oxidant status (TOS; in μmol H2O2 equivalents/l), oxidative stress index (OSI) and advanced oxidative protein products (AOPP; in μmol/L chloramine-T Eq). Serum TAS and TOS levels were measured using commercially available kits, and oxidative stress index (OSI) was calculated. Serum AOPP levels were evaluated using a colorimetric method. Peripheral blood lymphocytes were harvested and apoptosis level was determined via DNA ladder assay. Apoptotic, necrotic and live cell ratios stained with Acridine orange/Ethidium bromide (AO/EB) staining fluorescence method were calculated quantitatively through evaluation under a fluorescence microscope.
